Intracellular microcolonies of bacteria (IMC), in some cases developing large extracellular cysts (bacterial aggregates), infecting primarily gill and digestive gland, have been historically reported in a wide diversity of economically important mollusk species worldwide, sometimes associated with severe lesions and mass mortality events. As an effort to characterize those organisms, traditionally named as Rickettsia or Chlamydia-like organisms, 1950 specimens comprising 22 mollusk species were collected over 10 countries and after histology examination, a selection of 99 samples involving 20 species were subjected to 16S rRNA gene amplicon sequencing. The flat oyster Ostrea edulis L. is widespread along the Italian coasts. In particular, the Manfredonia Gulf (Adriatic Sea) represents an important site where natural beds subsist. Previous monitoring conducted in 1990 by light microscopy and ultrastructural studies revealed the presence of Bonamia-like microcell parasites in some flat oysters: following this observation, a new sampling of O. edulis was carried out at this location in 2007. The development of molecular diagnostic assays with increased sensitivity compared with conventional histological techniques is highly desirable for effective management of bonamiosis in cultured oyster stocks and wild populations. A real-time TaqMan PCR assay was developed for the specific detection of Bonamia species in infected oyster tissues. The TaqMan assay was shown to be significantly more sensitive than histopathology. Although a real-time TaqMan PCR assay is comparable with conventional PCR in terms of sensitivity, it offers the advantages that it is a rapid test and has a very low risk of sample cross-contamination. Nous disposons aujourd'hui d'o utils diagnostiques sensibles et spécifiques (PCR, HIS ... ) et d'un protocole de transmission de l' infection qui permettent d'assurer un suivi des productions larvaires en écloserie de bivalves pour les espèces considérées comme sensibles aux infections à herpèsvirus, mais également pour de nouvelles espèces. En effet, des mortalités massives (100 %) ont été rapportées dans une écloserie de coquille St-Jacques en Bretagne et ont pu être associées à la détection de OsHV -1. Les résultats obtenus concernant cette espèce, Pecten maximus, complètent et reflètent l'ensemble des travaux réalisés.

Le programme 4 portant sur la pathologie des coquillages dans le cadre d'une gestion durable des productions ostréicoles, comporte 2 actions principales réparties en une étude d'épidémiologie analytique et descriptive des agents pathogènes (Action 1), et l'étude des relations hôtes/agents pathogènes (Action 2). Les résultats 2001 présentés ici sont relatifs à ces 2 actions, avec notamment des informations sur les infections à virus de type herpès, et de recherche de bactéries pathogènes chez Crassostrea gigas lors des épisodes de mortalités estivales. Par ailleurs, l'émergence d'un programme national, intitulé MOREST (pour Mortalités Estivales) sur l' huître creuse C. gigas va permettre de rappeler le contexte scientifique des relations hôtes/pathogènes.

Etude du virus de type herpes observé chez les huîtres ArchiMer

Since 1991, sporadic, but high mortality in larvae of the Pacific oyster (Crassostrea gigas) has been observed during the summer in various French hatcheries. This mortality was concomitant with the detection of a virus by transmission electron microscopy. This virus, by virtue of its morphological characteristics and developmental cycle, seems to belong to the Herpesviridae family. The pathogenicity of this viral agent has been demonstrated in the laboratory by experimentally induced mortality on healthy larvae. Moreover, concomitant mortality has been reported in larvae of Crassostrea gigas and Ostrea edulis reared in the same tanks, with detection of a herpes-type virus.

Indeed, this unicellular parasite infects the flat oyster Ostrea edulis and multiplies within hemocytes, the central effectors of oyster defenses. Recent transcriptomic approaches focused on anti-viral immunity in molluscs lead to the assumption that the innate immune system, such as apoptosis, plays a crucial role against ostreid herpesvirus type 1 (OsHV-1), infecting Pacific cupped oyster, Crassostrea gigas. Since 2008, mass mortality outbreaks associated with the detection of particular variants of OsHV-1 have been reported in Crassostrea gigas spat and juveniles in several countries. Recent studies have reported information on viral replication during experimental infection. Viral DNA and RNA were also detected in the haemolymph and haemocytes suggesting that the virus could circulate through the circulatory system. Haplosporidiosis in the pacific oyster Crassostrea gigas from the French Atlantic coast ArchiMer

Two cases of haplosporidian infection occurred during 1993 in Pacific oysters Crassostrea gigas from the French Atlantic coast. The localization and ultrastructure of the plasmodia are described. In situ hybridization of infected tissue sections was conducted with DNA probes for oyster-infecting haplosporidians. The Haplosporidium nelsoni-specific DNA probe MSX1347 hybridized with the C. gigas parasite, and the H, costale-specific probe SSO1318 did not hybridize. Total genomic DNA was extracted from the infected tissue sections for polymerase chain reaction (PCR) amplification of the haplosporidian. PCR amplifications with H. nelsoni-specific primers and with 'universal' actin primers did not yield the expected products of 573 and 700 bp, respectively. A...
